加入角色专精设计：
除了补完剧情，《彩虹六号：维加斯2》另一个新特色是玩家能在剧情模式或是多人连线对战游戏中「专精」小队角色。利用一种称为「先进战斗及专精」（ACES）的系统，玩家将能强化人物角色在突击、狙击以及近距离战斗的技巧。

这些专精将与玩家的游戏风格息息相关，如果玩家偏爱把门轰开或是使用手榴弹，那么玩家在游戏中就会获得更多的炸药，如果喜欢神不知鬼不觉地用狙击枪把敌人乾掉，那么玩家就能得到威力更强的狙击步枪。也就是说，玩家越朝某项「专精技能」发展，就越能获得相关的强大武器。在新加入的11种新武器中， Ubisoft目前只透露有M468步枪和SR-25狙击步枪。

更灵活的小队AI：
采用强大的 Unreal Engine 3.0，玩家将看到更真实的罪恶之城，全新的场所、街道及小巷、工业区和脱衣舞俱乐部都将一一呈现在玩家眼前。游戏玩法承袭前作的「掩护及射击」机制，不过续作中将更为紧张刺激，因为掩护玩家的物体不再坚不可摧，像是木材或其他脆弱的掩体在炮火之下都会受到破坏。

因为加入这种设计，玩家的队友AI也重新编写过，让他们更灵活应对这种情况才能存活，举例来说，在前作中队友通常会朝着目标笔直前进，往往容易暴露在敌人的射线之中，在《彩虹六号：维加斯2》，由于掩体不再那么牢不可破，因此他们会采取一种称为「跃进」（Leap Frogging）的掩护技巧，一边采取不同位置的掩护，一边朝目标前进，不会在一个地方待太久，这种更真实的AI表现绝对会让玩家们感到大呼过瘾。
